Professor KIM Jin-Soo’s RNA-Guided Genome Editing Using Digenome-seq

Professor Kim Jin-Soo’s research team (Department of Chemistry) has announced Digenome-seq -- in vitro Cas9-digested whole-genome sequencing -- to profile genome-wide Cas9 off-target effects in human cells. Using Digenome-seq, the team successfully proved that CRISPR-Cas9, an RNA-guided genome editing system, works effectively and specifically on the target genome. The team also found that Cas9 off-target effects can be avoided by replacing 'promiscuous' single guide RNAs (sgRNAs) with modified sgRNAs. Professor Kim wrote that “Digenome-seq is a robust, sensitive, unbiased and cost-effective method for profiling genome-wide off-target effects of programmable nucleases including Cas9.” This research was published online on February 9 in Nature Methods.
